说明:CellG5检测试剂盒用于检测纤维素内切酶(1,4 -β-葡聚糖内切酶)包含两种组分:1) 4,6-O-(3-Ketobutylidene)-4-nitrophenyl-β-D-cellopentaoside (BPNPG5) 和 2)热稳定的β-葡萄糖苷酶。 酮阻断基团可阻止β-葡萄糖苷酶对BPNPG5的水解。通过纤维素内切酶的培养可生成非阻断的显色寡糖,被β-葡萄糖苷酶迅速水解。产生4-硝基苯酚的速率与β-葡萄糖苷酶水解BPNPG5直接相关。加入Tris缓冲液(pH 9.0)后可终止反应,并形成苯酚显色产物。

使用高纯度β-葡萄糖苷酶和苯亚甲基阻断,2 – 氯-4 – 硝基苯基-β-Dcellotrioside(BClPNPβ-G3)。 β-葡萄糖苷酶的能力确保测定的可靠的最大的灵敏度。BClPNPβ-G3的水解为苯亚甲基,通过纤维素酶阻断纤维二糖和2-氯-4 -硝基苯基-β-D-葡萄糖, 2-Cl-4-硝基苯基-β-D-葡萄糖通过β-葡萄糖苷酶立即裂解为D-葡萄糖和游离的2 – 氯-4 – 硝基苯酚(ClPNP)。

The CELLG3 assay reagent for the measurement of endo-cellulase (endo-1,4-β-glucanase) contains two components;

1) 4,6-O-benzylidene-2-chloro-4-nitrophenyl-β-D-cellotrioside (BCNPG3) and 2) thermostable β-glucosidase. The benzylidene blocking group prevents any hydrolytic action by the β-glucosidase on BCNPG3.  Incubation with an endo-cellulase generates a non-blocked colourimetric oligosaccharide that is rapidly hydrolysed by the ancillary β-glucosidase.  The rate of formation of 2-chloro-4-nitrophenol is therefore directly related to the hydrolysis of BCNPG3 by the endo-cellulase.  The reaction is terminated and the phenolate colour is developed on addition of Tris buffer solution (pH 9.0).
Please note that a new assay kit (K-CELLG5) is now available for the measurement of endo-cellulase.  The CELLG5 reagent contains a cellopentaose core and exhibits vastly improved sensitivity for some cellulases.  In addition, the exchange of the benzylidene blocking group in CELLG3 for 3-keto-butylidene in CELLG5 improves the substrate’s water solubility significantly, allowing for a reduction in the concentration of DMSO required in the assay.  As DMSO is known to inhibit certain cellulases, this is another benefit in using CELLG5.  Megazyme now recommends the use of K-CELLG5 for all assays for the measurement of endo-cellulase.
Colourimetric method for the determination of

      膳食纤维是一种多糖,它既不能被消化吸收,也不能产生能量。因此,曾一度被认为是一种“无营养物质”。后来发现了膳食纤维具有相当重要的生理作用。并被营养学界补充认定为第七类营养素,和传统的六类营养素——蛋白质、脂肪、碳水化合物、维生素、矿物质与水并列。

1.可溶性膳食纤维(来源于果胶、藻胶、魔芋等。魔芋盛产于我国四川等地,主要成分为葡甘聚糖)
2.不可溶性膳食纤维(全谷类粮食,其中包括麦麸、麦片、全麦粉及糙米、燕麦全谷类食物、豆类、蔬菜和水果等。)
  1. 简介:

膳食纤维是指碳水化合物及其相类似物质的总和,包括多糖、寡糖、木质素以及相关的植物物质。主要有纤维素、半纤维素、果胶及亲水胶体物质,如树胶、海藻多糖等组分;另外还包括植物细胞壁中所含有的木质素;不被人体消化酶所分解的物质,如抗性淀粉、抗性糊精、抗性低聚糖、改性纤维素、黏质、寡糖以及少量相关成分,如蜡纸、角质、软木脂等。
总膳食纤维检测试剂盒是根据AOAC991.43,AOAC 985.29,AACC32-07和AACC 32-05方法开发的,也适用于其它方法,如AACC Method 32-21,AACC method32-06。

  1. 检测原理

脱脂(如大于10%)干燥后的样品经热稳定a-淀粉酶、蛋白酶和淀粉葡萄糖苷酶酶解消化,酶解液通过乙醇沉淀、过滤,乙醇和丙酮洗涤残渣后干燥、称重,得到总膳食纤维(TDF)残渣;
酶解液通过直接过滤、热水洗涤残渣,干燥后称重,得到不溶性膳食纤维(IDF)残渣;
滤液用乙醇沉淀,过滤、干燥、称重,得到可溶性膳食纤维(SDF)残渣。
TDF、IDF和SDF的残渣扣除蛋白质、灰分和空白即得TDF、IDF和SDF含量。
该检测试剂盒的主要优势在于直接提供高纯度的酶,酶的活力是标准化的,而标准化的a-淀粉酶的活力在测量抗性淀粉是公认的。
淀粉葡萄糖苷酶中基本没有纤维素酶,而其他常用的酶制剂中常含有这种污染物,这将导致溶解和低估β-葡聚糖。
该试剂盒提供的酶都是稳定的液体,可以保存到试剂盒有效期,并且是直接应用液,不需要使用前再溶解。

  1. 用途

适用于检测谷物、水果、蔬菜和加工食品中的总膳食纤维(TDF)、不溶性
膳食纤维(IDF)和可溶性膳食纤维(SDF)的检测。

5.1 设备

  1. 高型无导流口烧杯:400mL或600mL
  2. 坩埚:具粗面烧结玻璃板,孔径40-60 μm。清洗后在马福炉中525℃灰化6小时或过夜,用真空泵除去硅藻土和灰分,室温下用2%清洗液浸泡1小时,用水和蒸馏水冲洗干净,用15 mL丙酮冲洗后风干,加1g硅藻土到干燥的坩埚中,在130℃干燥至恒重,在干燥器中冷却1小时,记下坩埚(包括硅藻土)的重量。

5.2 试剂

  1. 95%乙醇(体积比),
  2. 78 %乙醇:取821mL95%乙醇置于容量瓶中,用水稀释到刻度,混匀。
  3. 丙酮
  4. 蒸馏水或去离子水
  5. 清洗液Micro (International Products Corp.,Trenton,NJ).,配成2%液体
  6. MES/TRIS缓冲液(0.05mol/L):称取19.52g (MES) (Sigma, M 8250)和14.2 g

(TRIS) (Sigma,T1503),用1.7L蒸馏水溶解,用6mol/L氢氧化钠调节PH至
8.2±0.1,加水稀释到2L(注意24℃时PH值为8.2,20℃时PH值为8.3,27-28℃
时PH值为8.1)

  1. 盐酸溶液(0.561 mol/L):取93.5mL 6 mol/L的盐酸,加入700mL水中,混匀

后用水定容到1L。

  1. 氢氧化钠
  2. PH值标准缓冲液:PH值为 4.0,7.0 和10.0。

 

  1. 酶的纯化和标准化

6.1 根据AACC/AOAC的膳食纤维检测方法,必须确保酶的纯度。
6.1.1 淀粉葡萄糖苷酶被木聚糖酶、纤维素酶和果胶酶污染,会低估β-葡聚糖、
阿拉伯木聚糖和果胶的含量。
6.1.2 热稳定a-淀粉酶被污染,会影响抗性淀粉的测量。
6.1.3 蛋白酶被4-β-葡聚糖酶污染,会低估β-葡聚糖的含量。
6.1.4 如不是使用该酶,请参考下面方法测定。

6.2 根据AACC/AOAC的膳食纤维检测方法,必须对酶标准化测定,如果不是使用该的酶,请按以下方法测定
6.2.1 淀粉葡萄糖苷酶,0.2 mL,
— 酶活力表示方法1:淀粉/葡萄糖酶-过氧化物酶法,3300 U/mL,1个酶活力单位定义为:40°C,PH值4.5时,每分钟释放1 μmol葡萄糖所需要的酶量。
— 酶活力表示方法2:对-硝基苯基-b-麦芽糖苷(PNPBM)法,200 U/mL, 1个酶活力单位(1PNP单位))定义为:40°C,有过量萄糖苷酶存在下,每分钟从对硝基苯基-β-麦芽糖苷释放1 μmol 对硝基苯基所需要的酶量。
6.2.2 热稳定a-淀粉酶,0.05 mL,
— 酶活力表示方法1:以淀粉为底物,以Nelson/Somogyi还原糖表示,10000U/mL ,1个酶活力单位定义为:40°C,PH值6.5时,每分钟释放1 μmol葡萄糖所需要的酶量。
— 酶活力表示方法2:对硝基苯基麦芽糖为底物,3,000 U/mL(Ceralpha),1个酶活力单位定义为:40°C,PH值6.5时,每分钟释放1 μmol对-硝基苯基所需要的酶量。
6.2.3 蛋白酶,0.10 mL
— 酶活力表示方法1:酪蛋白测试,350 U/mL或50 mg/mL或7 Tyrosine U/mL,1个酶活力单位定义为:40°C,PH值8.0时,每分钟从可溶性酪蛋白中水解出(并溶于三氯乙酸)1 μmol酪氨酸所需要的酶量。
— 酶活力表示方法2:偶氮酪蛋白测试,350 U/mL或50 mg/mL或7 Tyrosine U/mL,1个内肽酶活力单位定义为:40°C,PH值8.0时,每分钟从可溶性酪蛋白中水解出(并溶于三氯乙酸)1μmol酪氨酸所需要的酶量。该偶氮酪蛋白(货号:S-AZCAS).
 

  1. 检测步骤

7.1 试样制备
准确称取双份试样各1 g,质量差小于0.005g,精确到0.1mg,置于400mL或600mL高型烧杯中,同时制备双份空白样,在每个烧杯中加入40 mL PH值8.2的MES-TRIS缓冲液,磁力搅拌,直到试样完全分散到缓冲液中。
7.2 热稳定a-淀粉酶酶解:加入50 μL热稳定a-淀粉酶溶液,加盖铝箔,置于95-100°C恒温振荡水浴中持续振摇,当所有烧杯全部放入水浴开始计时,反应35分钟。
7.3 冷却:将烧杯取出玲却到60°C,除去铝箔盖,用刮勺将烧杯内壁和底部的胶状物刮下,用10 mL蒸馏水冲洗烧杯壁和刮勺。
7.4 蛋白酶酶解:在每个烧杯中各加入100 μL蛋白酶溶液,加盖铝箔,置于60°C恒振荡水浴中,当烧杯内温度达到60°C开始计时,持续反应30分钟。
7.5 PH值调节:反应30分钟后,边搅拌边加入0.561 mol/L盐酸,严格控制60°C,用 1mol/L氢氧化钠溶液或 1mol/L盐酸溶液,控制PH值在4.5±0.2。
7.6 淀粉葡萄糖苷酶酶解:在上述溶液中边搅拌边加入200 μL淀粉葡萄糖苷酶溶液,加盖铝箔,置于60°C恒温振荡水浴中,持续振摇,当烧杯内温度达到60°C开始计时,反应30分钟。

  1. 测定

8.1 不溶性膳食纤维测定:
8.1.1 过滤洗涤:试样酶解液全部转移到坩埚中过滤,残渣用10mL预热到70°C的蒸馏水洗涤两次,合并过滤液,转移至另一600mL高脚烧杯中,备测可溶性膳食纤维。残渣分别用10mL 95%乙醇和丙酮洗涤两次,抽滤去除洗涤液,将坩埚连同残渣在103°C烘干过夜,将坩埚置于干燥器中冷却1小时,称重(包括坩埚、膳食纤维残渣和硅藻土),精确至0.1mg,减去坩埚和硅藻土的干重,计算残渣质量。
8.1.2 蛋白质和灰分测定:称完质量的残渣和硅藻土的混合物,一份用凯氏定氮法,以N×6.25为换算系数,计算蛋白质质量。另一份在525°C灰化5小时,于干燥器中冷却,精确称量坩埚总量(精确至0.1mg)减去坩埚和硅藻土干重,计算灰分质量。
8.2 可溶性膳食纤维测定
8.2.1 计算滤液体积:将不可溶性膳食纤维过滤后的滤液收集到600mL的高型烧杯中,通过烧杯+滤液总重扣除烧杯质量的方法估算滤液的体积。
8.2.2 沉淀:将滤液加入4倍体积预热60°C的95%乙醇,或取80g滤液加入320mL预热60°C的95%乙醇室温下沉淀1小时。
8.2.3 过滤:在干燥的坩埚中加入1g硅藻土,70°C真空干燥至恒重,记录坩埚的质量(精确到0.1mg)。用15mL 78%乙醇将硅藻土润湿,并用真空溶剂过滤装置在抽真空条件下,使硅藻土平铺于坩埚中。将样品酶解液缓慢转移至对应的坩埚中,抽滤。用刮勺和78%乙醇将所有残渣转至坩埚中。
8.2.4 洗涤:分别用15mL 78%乙醇、15mL 95%乙醇和15mL丙酮洗涤残渣各两次,抽滤去除洗涤液后,将坩埚连同残渣在105°C烘干过夜。将坩埚置于干燥器中冷却1小时,称重(包括坩埚、膳食纤维残渣和硅藻土),精确至0.1mg。减去坩埚和硅藻土的干重,计算残渣质量。
8.2.5 蛋白质和灰分测定:称完质量的残渣和硅藻土的混合物,一份用凯氏定氮法,以N×6.25为换算系数,计算蛋白质质量,另一份在525°C灰化5小时,于干燥器中冷却,精确称量坩埚总量(精确至0.1mg)减去坩埚和硅藻土干重,计算灰分质量。
8.3 总膳食纤维检测检测
8.3.1 沉淀:在每份试样中加入预热至60°C的95%乙醇225mL(预热后的体积,如乙醇的温度超过65°C,则加入228mL),乙醇与样液体积比为4:1,取出烧杯,盖上铝箔,室温下沉淀1小时。建议改为:称量酶解液的质量,用天平加入4倍质量的预热的60°C的95%乙醇,4°C冰箱中沉淀过夜。
8.3.2 过滤:在干燥的坩埚中加入1g硅藻土,70°C真空干燥至恒重,记录坩埚的质量(精确到0.1mg)。用15mL 78%乙醇将硅藻土润湿,并用真空溶剂过滤装置在抽真空条件下,使硅藻土平铺于坩埚中。将样品酶解液缓慢转移至对应的坩埚中,抽滤。用刮勺和78%乙醇将所有残渣转至坩埚中。

  1. 氢氧化钠溶液(0.275mol/L):去11g氢氧化钠溶解在700mL蒸馏水中,冷却转

移到容量瓶中,用水定容到1L。

  1. 盐酸溶液(0.325 mol/L):取 325 mL 1.0 mol/L 盐酸置于容量瓶中,用水定容

到1L。

  1. 样品准备

总膳食纤维的测定必须是低脂或无脂的干燥样品,取混匀后的样品于70°C真
空干燥过夜,干燥器中冷却,再称重记录重量损失,干样粉碎后过0.3-0.5 mm
筛,若试样不能加热,则冷冻干燥后再粉碎过筛。如果是高脂肪样品(> 10 %),
取经过干燥的样品分别用25mL石油醚脱脂三次,干燥后记录脱脂的质量损失,
最后计算总膳食纤维含量时进行进行校正,粉碎过筛后的干燥试样放在干燥
器中待用。

  1. 分析步骤

准确称取双份试样各1 g,质量差小于0.005g,精确到0.1mg,置于400mL或
600mL高型烧杯中,同时制备双份空白样,在每个烧杯中加入50 mL PH值6.0
的磷酸盐缓冲液,磁力搅拌,直到试样完全分散到缓冲液中,控制PH值6.0±0.1。
4.2 热稳定a-淀粉酶酶解:加入50 μL热稳定a-淀粉酶溶液,加盖铝箔,置于沸腾
水浴中15分钟,每5分钟轻轻振荡一次。当烧杯内的温度到100°C开始计时,
总共大约30分钟。
4.3 冷却:将烧杯取出冷却到室温,加入10 mL 0.275mol/L 的氢氧化钠,调节
PH值在7.5±0.1。
4.4 蛋白酶酶解:在每个烧杯中各加入100 μL蛋白酶溶液,加盖铝箔,置于60°C
恒振荡水浴中,当烧杯温度达到60°C开始计时,持续反应30分钟。
4.5 冷却:加入10 mL 0.325 mol/L盐酸,调节PH值在4.5±0.2。
4.6 淀粉葡萄糖苷酶酶解:在上述溶液中边搅拌边加入200 μL淀粉葡萄糖苷酶溶
液,加盖铝箔,置于60°C恒温振荡水浴中,持续振摇,当烧杯内温度达到60°C
开始计时,反应20分钟。
4.7 在每份试样中加入预热至60°C的95%乙醇280mL(预热前的体积),乙醇与样
液体积比为4:1,取出烧杯,盖上铝箔,室温下沉淀1小时。
4.8 过滤:在干燥的坩埚中加入1g硅藻土,70°C真空干燥至恒重,记录坩埚的质
量(精确到0.1mg)。用15mL 78%乙醇将硅藻土润湿,并用真空溶剂过滤装置
在抽真空条件下,使硅藻土平铺于坩埚中。将样品酶解液缓慢转移至对应的
坩埚中,抽滤。用刮勺和78%乙醇将所有残渣转至坩埚中。
4.9 洗涤:分别用20mL 78%乙醇洗涤3次、10mL 95%乙醇和10mL丙酮洗涤残渣
各两次,抽滤去除洗涤液后,将坩埚连同残渣在105°C烘干过夜或70°C真空
干燥过夜。将坩埚置于干燥器中冷却1小时,称重(包括坩埚、膳食纤维残渣
和硅藻土),精确至0.1mg。减去坩埚和硅藻土的干重,计算残渣质量。
4.10 蛋白质和灰分测定:称完质量的残渣和硅藻土的混合物,一份用凯氏定
氮法,以N×6.25为换算系数,计算蛋白质含量。另一份在525°C灰化5小时,
于干燥器中冷却,精确称量坩埚总量(精确至0.1mg)减去坩埚和硅藻土干重,
计算灰分质量。

     β-葡聚糖酶作用方式不同,可分为内切型和外切型。前者存在于谷物种子、某些真菌和某些细菌中,能催化水解谷物细胞壁中的β-葡聚糖,其中包括内切型β-1,4-葡聚糖酶、内切型β-1,3-葡聚糖酶。后者存在于谷物种子中,其中又包括外切型β-1,4-葡聚糖酶、外切型β-1,3-葡聚糖酶。
在工业上采用细菌(如枯草芽孢杆菌)、曲霉(如黑曲霉)、青霉(如伊氏青霉)生产。在啤酒生产原料大麦及其麦芽中,含本品较多。在啤酒生产中采用β-葡聚糖酶,可提高麦汁的过滤速度和得率,从而保证啤酒质量。也可用于饴糖、麦芽糖浆生产和用作饲料添加剂。

许多食品和饮料中都发现有D-乳酸.  由乳酸菌产生, D-乳酸存在于牛奶的发酵产品中比如酸奶,腌菜,腌肉和腌鱼中。牛奶、肉、和果汁的质量可以通过测定D-乳酸的含量来确定。在葡萄酒生产中,D-乳酸的产生标志着酒被乳酸菌***。在化学工业中,D-乳酸和L-乳酸都是生产过程中聚合物的原料,比如聚交酯和一些生物学上可降解的聚合物。同时也应用在化妆品和医药品中。
原理:
D-乳酸的定量分析需要两步酶化反应。  第一个反应由D-乳酸脱氢酶(D-LDH)催化,D-乳酸在烟酰腺嘌呤二核苷酸(NAD+)的催化下,被氧化成丙***酸
然而, 因为反应(1)的移动和D-乳酸和NAD+有着密切的关系,下一步反应需要消耗掉产生的丙***酸 在大量D-谷氨酸的存在下,通过在D-谷氨酸-丙***酸转氨酶(D-GPT)的作用下把丙***酸转化为D-丙氨酸和2-***戊二酸。(2).
以上两个反应中产生的 NADH 的量可以用来计量D-乳酸的量。  NADH的量通过340nm处增加的吸光率来测量。
特异性,敏感度, 线性和准确性:
此方法特异性检测D-乳酸.
此方法特异性检测D-乳酸和L-乳酸。在D-乳酸锂(MW=96.0)的检测中,大约可以预期结果为96%(W/W),L-乳酸锂可以得到98%(W/W)。
检测的灵敏度为0.005 个吸光度单位。相当于样品最大体积为1.50 mL 时,样品浓度为0.107 mg/L(或者 1.60 mg/L ,样品体积0.1 mL) 。检测限为 0.214 mg/L,这是在0.010的吸光度差异,样品最大体积为1.50 mL为得到的 。
D-乳酸检测的线性变化为0.5 到 30 μg 。在重复测定中,同一份样品溶液,可能产生0.005 到 0.010 个吸光光度值的差异。这就相当于样品1.50 mL,样品溶液中D-乳酸的浓度为0.107 到 0.214 mg/L 。如果样品在制备阶段被稀释过,结果要乘以稀释倍数F。如果样品制备阶段,称量过样品,比如10 g/L,可能会产生0.02 到 0.05 g/100 g 的差异。
干扰:
如果D-乳酸的转换在实验指定的时间内(大约5分钟)彻底完成,通常认为是没有发生任何干扰。不过,这个也可以进一步验证,通过在实验的完成时,向比色皿中加入D-乳酸混合物(大约每0.1mL15ug),吸光光度值有会明显增加。
样品中的干扰物质的分析可以通过加入一个内参来确定。需要进行一个标准品定量回收实验,通过这个回收实验(也就是在最初的提取步骤中向样品中加入D-乳酸)可以算出样品在处理和提取过程中的损失量。
