SMARTer Mouse TCR a/b Profiling Kit 为那些寻求对T细胞受体（TCR）库进行NGS分析的研究者提供了一种强大的新的解决方案。试剂基于5’-RACE 方法可以捕获TCR转录本完整V(D)J 可变区，可以低至10 ng 至500 ng的来源于小鼠脾脏、胸腺或PBMCs的总RNA起始，也可以1,000 至 10,000个纯化的T细胞起始。顾名思义，本试剂可以在同一实验或单独实验中获得α和β链链多样性数据。
与采用多重PCR方法研究TCR多样性不同，每轮文库扩增只使用针对不同TCR亚基的一对引物扩增，提高了效率，减少了引物二聚体形成的可能性。基于多重PCR的分析方法，众所周知容易形成与引物相关的序列特异性偏差；这些偏差在我们基于RACE的方法中不会存在，因为所有的可变区都由相同的恒定区序列引发。采用本试剂~4 h即可构建indexed文库用于Illumina平台测序。
试剂采用了SMART (Switching Mechanism at 5’ End of RNA Template) 技术和LNA技术，可以无偏扩增TCR mRNA序列，并具有很高的灵敏度。

SMARTer Mouse BCR IgG H/K/L Profiling Kit—捕获BCR转录本完整V(D)J 可变区

B 细胞是适应性免疫应答的重要组成部分，其表面表达B细胞受体（BCR），可以识别病原体中的特定分子模式。了解BCRs图谱（也就是由特定H,K,L基因片段构成的受体和克隆型多样性）不仅有助于深入了解健康个体中的适应性免疫应答，也有助于了解多种疾病的适应性免疫应答。准确确定免疫系统表达的克隆型和同种型有助于全面了解B细胞群体。
 
新推出的SMARTer Mouse BCR IgG H/K/L Profiling Kit基于5’RACE和NGS技术，提供了一种灵敏，准确，优化的BCR库分析方法。虽然采用多重PCR可以在一个反应中对多个BCR基因进行扩增，但也被证明在对特定序列进行扩增时存在灵敏度，特异性和偏差的问题，这些都可能带来同种型的鉴别困难。而5’RACE方法减少了变异性并且可以从BCR重链或轻链的恒定区引发扩增。试剂盒将这些优点与基因特异性扩增相结合可以捕获BCR完整V(D)J可变区，提供了一种高灵敏度、可重复的B细胞库分析方法。在大部分情况下，通过H/K/L链的下游测序分析可以准确鉴别克隆型，可靠确定同种型。

祖细胞发育过程中胚系基因片段经过V,D,J重排形成两条相同的重链。V,J片段重排形成两条相同的轻链。V,D,J片段连接处的核苷酸随机插入或缺失增加了额外的多样性。B细胞经过免疫应答刺激后通过体细胞高频突变（SHM）进一步引入更多的点突变。

PanelA dT-引物起始合成一链cDNA，经过两轮PCR扩增合成cDNA文库。PCR产物经纯化，片段筛选和质量检测后用于测序分析。
PanelB对于每个样品，在样品中所有mRNA 进行RT反应后，使用者可以通过两轮基因特异性巢式PCR特定扩增IgG 的1-3条链。

对于每一个样品，在所有mRNA进行RT后，使用者可以特定扩增IgG的1-3条链。每个扩增反应需要5μl RT产物。第一轮PCR后，各取1μl PCR反应产物用于独立PCR反应，分别扩增每条链，同一样品添加相同的indexes，不同的样品添加不同的indexes。经过PCR 2最终扩增后，使用者可以通过Bioanalyzer 或Fragment Analyzer分析每个反应产物，可以以此选择哪条扩增链进行混合测序。

以来自于不同IgG亚型的自有杂交瘤（10E8）和两个ATCC杂交瘤样品（HB-8117和TIB-127）的10 ng RNA起始，采用SMARTer Mouse BCR IgG H/K/L Profiling Kit构建包含BCR重链和轻链（kappa）的文库。Panel A Bioanalyzer结果显示了每个杂交瘤重链和轻链的基因特异性扩增结果。在每个样品中都观察到了700-900 bp的预期峰值。电泳图中明显的峰表明在这些杂交瘤中表达kappa链。相反，lambda链测序库相对较宽并且大小低于预期。每个样品中标注为“LM”和“UM”的峰为DNA reference marker。Panel B 采用MiXCR软件（version 1.8）确定比对指标，与所有IG序列进行了比对分析。MiXCR软件对前2个克隆的分析结果显示了V,D,J等位基因及通过软件分析确定的同种型和亚类。

It does not cross-react with full length caspase-9 or with other caspases at endogenous levels.Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Asp353 of mouse caspase-9. Antibodies are purified by protein A and peptide affinity chromatography.
