鲁米诺(luminol)，又名发光氨。化学名称为3-氨基邻苯二甲酰肼。常温下是一种黄色晶体或者米黄色粉末，是一种比较稳定的人工合成的有机化合物。鲁米诺是通用的发光化学试剂，与适当的氧化剂混合时会发出引人注目的蓝色光。它是白色至淡黄色的晶体，可溶于水和大多数有机极性溶剂，是一种HRP过氧化酶发光性底物。金畔生物是AAT Bioquest的中国代理商，为您提供最优质的鲁米诺(3-氨基邻苯二甲酸酐)。

因dUTP的浓度设定是dTTP的3倍，所以总体dNTP的浓度变高，虽然PCR Buffer中含有MgCl2，但为了保持MgCl2与dNTP量的平衡，本制品中的Mg2+浓度高于TaKaRa Taq Hot Start Version (Code No. R007A)附带的10× PCR Buffer (Mg2+ plus)。

■ 制品说明
PCR是一种非常敏感的DNA扩增技术，易受污染影响，PCR扩增产物的污染会产生假阳性结果。PCR扩增产物的污染是End-point PCR方法，特别在食品、环境检测中是令人头痛的问题。
本制品是一种可防止假阳性产生的试剂盒。使用本制品是在PCR反应时以dUTP取代dTTP，并同时使用Hot Start型DNA聚合酶TaKaRa Taq HS进行PCR扩增反应。反应液配制后，对含有dU的DNA具有降解作用的UNG酶（Uracil-N-glycosylase）可选择性水解含有尿嘧啶的PCR产物，而对不含尿嘧啶的模板无任何影响。
UNG的作用原理：在PCR反应前25℃、10分钟反应中，UNG酶可水解PCR反应液中含有尿嘧啶的PCR产物的尿嘧啶碱基和糖磷酸骨架的N-糖苷键，产生脱嘧啶位点。随后进行95℃、2分钟热处理，在UNG酶失活的同时进一步对磷酸骨架水解，从而消除含有尿嘧啶的PCR产物的污染。
UNG酶可以作用于单链或双链DNA，对RNA无活性。

■ 注意事项
从反应液的配制到PCR产物的检出应分别在4个操作区操作，操作区推荐进行物理性隔离。
操作区1：反应液的配制及分装。
操作区2：样品的制备。
操作区3：将样品添加到反应液中。
操作区4：PCR反应及凝胶电泳对PCR产物进行检出。
为防止污染，除在操作区4外，请避免对装有PCR扩增产物的tube进行开闭。

    Cy3 酪胺是美国AAT Bioquest生产的花菁染料，对于许多免疫组织化学（IHC）应用，传统的酶促扩增程序足以实现足够的抗原检测。但是，有几个因素限制了这些程序的敏感性和实用性。事实证明，酪胺信号放大（TSA）是一种特别通用且功能强大的酶扩增技术，具有更高的检测灵敏度。 TSA基于HRP在低浓度过氧化氢存在下将含有标记酪胺的底物转化为氧化的高反应性自由基的能力，该自由基可与HRP处或附近的酪氨酸残基共价结合。为了实现最大的IHC检测，酪胺用荧光团预标记。每个过氧化物酶标记物的多个酪胺底物的转换所赋予的信号放大转化了对低丰度靶标的超灵敏检测以及使用较少量的抗体和杂交探针。在免疫组织化学应用中，源自TSA方法的灵敏度增强允许增加一抗稀释以减少非特异性背景信号，并且可以克服由次优固定程序或低水平靶表达引起的弱免疫标记。 Cy3酪胺含有明亮的Cy3，可以使用标准Cy3过滤器组轻松检测。金畔生物是AAT Bioquest的中国代理商，为您提供最优质的Cy3 酪胺检测试剂。
