◆产品描述
Dectin-1和dectin-2是C型凝集素家族的II型跨膜蛋白。 它们在其细胞外的区域都有单个糖基识别结构域(CRD)。 Dectin-1和Dectin-2在巨噬细胞和树突状细胞中表达。Dectin-1通过其胞浆中的免疫受体酪氨酸抑制基序(ITAM)样基序及其CRD来识别β-葡聚糖与信号,而dectin-2则识别α-甘露聚糖并通过与含有ITAM的Fc受体γ-链结合来转导其信号。Dectin-1通过检测β-葡聚糖并通过增加Th17介导的免疫应答来控制真菌感染。 Dectin-1似乎也能检测到非真菌配体,如无脊椎动物原肌球蛋白。Dectin-1通过结合非真菌配体,来调控IL-33驱动的天然淋巴细胞产生的IL-13,从而控制肺中由尘螨空气过敏原引起的2型免疫应答。

气道祖细胞/肺泡祖细胞标记物
抗CPM,单克隆抗体(WK), 结合红色荧光染料(635)
羧肽酶M(简称CPM)是出现在细胞膜表面的肽链外切酶。它从肽和蛋白质催化裂解出C端精氨酸和赖氨酸残基。近年研究报道,它可作为气道祖细胞、肺泡祖细胞、肝祖细胞的标志物。此产品为红色荧光染料(635)标记处理后的抗CPM单克隆抗体(WK)。此产品专用于流式细胞术,无需二次抗体染色。
◆应用案例
将人类iPS细胞分化为肺祖细胞。细胞分散后,使用此新开发的产品进行抗体染色。
通过流式细胞仪进行分类。分选后,对细胞群进行免疫染色。

数据提供:京都大学大学院医学研究科 后藤真平教授
结果:发现其中高表达CPM的细胞群同时高表达NKX2-1。
结果:NKX2-1是呼吸细胞的标志物。

研究背景:前列腺素 E2(PGE2)由于其在炎症、癌症、动脉粥样硬化、自身免疫性疾病和脓毒症中具有重要地位而被广泛研究。氧化的花生四烯酸通过前列腺素合成酶(COX-1和COX-2)生成前列腺素 H2(PGH2),被前列腺素合成酶进一步代谢成主要产物前列腺素 E2(PGE2)。前列腺素 E2(PGE2)通过与细胞表面的 G-蛋白偶联受体(EP1、EP2、EP3 和 EP4)的结合来介导自分泌和旁分泌的信号传导,从而调节磷脂酶C和腺苷酸环化酶活性。PGE2 作为一种治疗靶点,无论是通过 COX 抑制剂(NSAIDS)调节其合成,还是通过下调其与受体结合或结合拮抗剂,都引起了人们极大的兴趣。在各种组织中产生的前列腺素 E2(PGE2)已在多种生理过程中表现出了调节作用,包括在肾脏中尿钠排泄,血管中平滑肌的弹性,以及单核细胞和巨噬细胞对损伤组织的炎症作用。
