Bcl10 Antibody (331.3) is a high quality monoclonal Bcl10 antibody (also designated BCL10 immune signaling adaptor antibody) suitable for the detection of the Bcl10 protein of mouse, rat and human origin. Bcl10 Antibody (331.3) is available as both the non-conjugated anti-Bcl10 antibody form, as well as multiple conjugated forms of anti-Bcl10 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Bcl10, also designated CIPER, c-CARMEN and mE10, was first identified as a gene truncated or mutated in MALT B cell lymphomas and other tumor types. Bcl10 is homologous to the equine herpesvirus-2 E10 gene and, like E10, it contains an N-terminal caspase recruitment domain (CARD). Expression of Bcl10 has been shown to induce NFκB activation in a NIK-dependent pathway, and research indicates that the CARD domain is essential for this activation; although in a separate study, Bcl10 by itself did not induce JNK or NFκB activation. Overexpression of Bcl10 has been shown to induce apoptosis in a manner dependent on CARD-mediated oligomerization. Bcl10 has also been shown to play a role in processing of caspase-9 to its active dimer. Other studies have shown that Bcl10 is not mutated in many human tumors and lymphomas.

PicoPLEX Single Cell WGA Kit v3 (PicoPLEX WGA v3)可以从单细胞扩增DNA，获得高度可重复的文库。细胞裂解和预扩增后，进行低背景的PCR扩增，并在3小时内获得微克级产物。

以单细胞 (1cell) 和5个细胞 (5 cells) 的GM12878细胞系 (Coriell研究所) 起始 (一式两份)，分别使用PicoPLEX WGA v3,A公司试剂盒和B公司试剂盒，以及bulk gDNA对照制备全基因组扩增产物，进行SNV检测的比较。其中B公司试剂盒虽然制备了足够的扩增产物，但只有5 cells样本中一个包含足够的扩增子进行测序，而其他样本无可用数据进行测序。根据Genome In a Bottle (GIB) 和hg19 (human Genome assembly GRCh37, Ensembl) 数据库的交叉分析，预计GM12879细胞系总共存在78个SNVs。由于扩增子的设计，2×75 bp的读取长度很短，78个SNVs中的4个未能检测到。因此，检测的SNV总数减少到74个。使用VarDict软件，以≥10reads (10X coverage) 和等位基因频率≥20％为基准从BAM文件分析SNV。与其他两个公司试剂盒相比，PicoPLEX WGA v3具有更高的call rates和更低的等位基因丢失率。

以来自不同细胞系(GM22601、GM05067和GM12878)的单细胞起始，使用PicoPLEX Single Cell WGA Kit v3扩增基因组。之后取1ng的扩增产物，使用Illumina Nextera XT Kit制备文库，并在Illumina MiSeq（PE 75）进行测序。对Fastq文件去除接头序列信息后，与human genome assembly GRCh37进行比对。只分析常染色体。对于这两个panels，校准归一化至100万reads，使用bedtools 2.25.0计算每1 Mb的 bin的reads数。使用Integrative Genomics Viewer (IGV)绘制bin counts的log2 ratio (Sample/Reference)。PicoPLEX Single Cell WGA Kit v3还显示了可以高品质的进行染色体异倍性分析。
