中和抗体（Nab）有着可通过结合抗原、中和病原体来阻止病原体感染靶细胞的特点，与其结合后的病原体会变为非传染性。

COVID-19特异性中和抗体Nabs可与病毒表面的刺突蛋白，即S蛋白结合，使COVID-19病毒无法与人细胞受体ACE2结合。这个过程被称为“免疫消毒”，免疫系统在病毒感染人体细胞之前就消灭了传染性颗粒。

中和抗体NAbs的产生是感染COVID-19或接种COVID-19疫苗时所必要的免疫反应，是社会获得群体免疫的基础。

为什么我们需要检测中和抗体？

l 衡量疫苗接种的效果（针对COVID-19的任意疫苗）

l 衡量社会的免疫力

l 寻找抗血清捐赠者（捐赠给ICU重症患者）

一般来说，检测中和抗体Nabs，需要使用蚀斑减少中和试验（PRNT），这是一种具有感染性的方法，需要在高安全性实验室S3中进行。而这款COVID-19中和抗体ELISA试剂盒，实验步骤得到简化，无需任何预防措施。该测试可用于一般自动化检测，只需90 min即可得出检测结果。

◆检测步骤

 样品制备

 将90 μL样品稀释液和10 μL血清/血浆加入EP管或预混板中；

 添加100 μL对照品；若是定量检测，则添加100 μL标准品至EP管或预混板；

 将100 μL的RBD偶联物添加至稀释的样品、对照组和标准品中；

 混合均匀。

 预孵育

 在37°C下孵育30±5 min

 样品转移和分析、孵育

 将100 μL经孵育溶液从EP管或预混板转移到ACE2蛋白包被的微量滴定板的孔中；

 在37°C下孵育30±5 min

 洗涤

 以300 μL/孔洗涤3次

 添加/孵育 

 底物溶液

 底物溶液：100 μL/孔；

 在37°C孵育15±2 min

 终止反应

 加入终止液：50 μL/孔；

 检测
